Peptide Fractionation

Proteomes are too complex to completely profile in a single analysis so achieving broad and deep coverage often necessitates a pre-fractionation step. We have tried lots of ways to do this and have some favorites. Drop by for a day or two and we’ll walk you through them.

Protein Digestion

Shotgun proteomics relies on the analysis and detection of peptides – each ~6 to 20 amino acids in length – created following enzymatic proteolysis of intact proteins.  Incomplete and/or semi-specific digestion can really dampen your results.  If you are having issues with digestion we can assist by sharing with you our protocols and helping you assess the quality of your digestion.

Protein Extraction

Proteome analysis requires homogenization and extraction of proteins from all other cellular components.  The methods employed to accomplish these tasks are dependent upon sample type.  We list here the various sample types we have experience working with.
